Hi, Jennifer,
Every file of GIS coordinates is written with respect to some projection. That should be determined and set before the data is brought into ArcMap. If a shapefile has no .prj file, then one should be created for it by editing it with ArcCatalog.
Here are some instructions:
Once the shapefile has a projection defined, ArcMap can reproject as necessary to the current map view, as defined by the data frame (“Layers”).

I'm with Jarlath, either a data layer is defined incorrectly, or when you defined a projection it was not the correct projection for the data.

I'm trying to help out a colleague. He has a map that has data coming in at three different areas even when I've done define projection, reproject, open a clean mxd and define the map projection before loading data, but I am out of ideas. We've gone through a few transformations as well, but y'all know how many of those there are. Can I provide any more information or data (limited) to get some more help?
